Ubisoft today detailed its plans to release Assassin’s Creed Brotherhood DLC exclusively for the PlayStation3. The Copernicus Conspiracy will be available for free via the PlayStation Network and features several new missions from the famous Renaissance astronomer Copernicus.
Ezio will be tasked with foiling the conspiracy against Copernicus and defending his philosophy as he embarks on a variety of new courier, assassination and protection missions.
The Copernicus Conspiracy will be available on November 19th.

My guess is a sequel to the PSP game in 2011, then the final PS3/Xbox360 game in 2012 (as people keep saying, for obvious reasons).
Based on no evidence though, just a hunch.

The story certainly spins-off from from ACII, but the city of Rome is new and there are a lot of added features in the single-player game that weren't in ACII. It's effectively Assassin's Creed 2.9.
